The NFL's rushing leader suffered an oblique injury and was reported as questionable to return.
He attempted to play through it but headed off the field after a run that was called back by a penalty.
McCaffrey then headed to the locker room, indicating that his day is likely done.
With two of the most versatile and dangerous players in the NFL ruled out, the 49ers lose much of what makes their offense special, the gravity of both McCaffrey and Samuel continually causing myriad problems for opposing defenses.
